Laboratory for Stem Cell Transplantation and Immunotherapy

Our center is specially focused on haplo (=half) identical stem cell transplantation. This procedure offers the possibility for parents to serve as stem cell donors for their children. Complex purification of stem cell products is necessary in order to avoid severe graft rejection due to half compatibility of donor and recipient.

Manufacturing of our cell products is performed under GMP-conditions by German Red Cross Donor Service Baden-Württemberg-Hessen (BSD) according to the German Medicine Act (German drug law, AMG). This is done in close cooperation with our laboratory and our physicians.

In order to realize the transplantation from autologous, allogeneic and especially haploidentical stem cell products we are performing several modern purification procedures using CliniMACS technology in special cleanrooms and in the context of the manufacturing authorization of BSD.


        Being a delegated testing laboratory of BSD (Test activity according to §14 Paragraph 4 No. 4 AMG) our focus is on quality control of these stem cell preparations. In accordance with strict drug law regulations we determine cell amounts by flow cytometry.

          QC Determination

            Our laboratory has more than 10 years’ experience in this area and is certified according to DIN EN ISO 9001:2008; accredited according to DIN EN ISO 15189 (Certificate) and accredited according to JACIE (Certificate).

            Furthermore several cellular immunotherapies are applied in our center – especially tumor- and leukemia specific T-cells, NK cells and cytokine-induced killer (CIK) cells.

            In March 2016 a multicenter prospective clinical Phase I/II Study (EudraCT number 2013-005446-11) investigating the clinical usage of cytokine-induced killer (CIK) cells in patients with high-risk leukemia or myelodysplastic syndromes and expected relapse after allogeneic stem cell transplantation has started.

            As well as the stem cells, cellular immunotherapies are manufactured at BSD; our laboratory serves as a delegated testing laboratory likewise.

                1) Quality control of stem cell preparations and immunotherapies

                Products for stem cell transplantation:

                • CD3/CD19-depletion
                • Bone marrow and unmanipulated leukapheresis products
                • Autologous stem cell products

                Cellular immunotherapies:
                • Cytokine-induced killer cell infusions (CIK)

                2) Development of new stem cell transplantation and quality control procedures and immunotherapies/translational Research

                To optimize and develop new therapies we perform targeted research at the pre-clinical and clinical interface. Special topics in the field of our translational research are:

                Development of cellular immunotherapies - „from bench to bedside“:
                • Cytokine-induced killer cells (CIK)
                • CIK-cell-based therapy against virus infections
                • Tumor- and leukemia-specific T-cells
                • NK-cells

                3) Immunophenotyping after stem cell transplantation / Immunodeficiency diagnostics

                Evaluation of cellular immune status in patients after stem cell transplantation or children with suspected immunodeficiency is an additional focus of our laboratory.
                Complex characterization of immune cells and their function is performed in patients after stem cell transplantation (immune reconstitution), in patients with recurrent infections and for diagnosis of primary or secondary immunodeficiency.

                  (CD3+CD4+ Helper T-cells, CD3+CD8+ cytotoxic T-cells and their subgroups: activated, naïve, effector memory (TEM), central memory (TCM), effector memory RA+ (TEMRA), TCRαβ, TCRγδ, regulatory T-cells (Treg), CD4+CD8+ double-positive and CD4CD8 TCRαβ+ double-negative T-cells)
                  (naïve, switched, non-switched, memory B-cells)
                  (CD3CD56+ NK-cells and their subgroups: CD3CD56brightCD16 immunregulatory, CD3CD56dimCD16+ cytotoxic NK-cells)
                  (plasmacytoid and myeloid dendritic cells, monocytes)

                      • Functional tests
                      Granulocyte function test (oxidative burst)

                      • Cell typing in bronchioalveolar lavage fluid, liquor und ascites

                      • CD34 stem cell monitoring after G-CSF stimulation
